Reverzna transkriptaza je encim (EC 2.7.7.49), ki spada v razred transferaz. Katalizira podaljševanje DNA verige na 3'- koncu na osnovi RNA matrice, torej omogoča prepis RNA na DNA (en. RNA-directed DNA polymerase). Za začetek potrebuje že obstoječ RNA ali DNA primer – ne more ustvariti verige od začetka ("de novo" ).[1] Prepis poteka tako, da na obstoječi primer dodaja posamezne nukleotide, ki so komplementarni RNA matrici, torej katalizira proces, ki poteka v nasprotni smeri kakor transkripcija

Odkrila sta jo neodvisno drug od drugega Howard Temin, University of Wisconsin-Madison in David Baltimore, MIT (Massachusetts Institute of Technology). Skupaj z Renatom Dulbeccom so za ta dosežek leta 1975 prejeli Nobelovo nagrado.
