Barnaza namreč nima disulfidnih vezi, niti ne zahteva divalentnih kationov ali nepeptidnih komponent za to, da se lahko veže. Zaradi te preprostosti v kombinaciji z reverzibilnim harmoničnim prehodom, je bila barnaza izredno natančno preučena, predvsem da bi razumeli kako se proteini povezujejo. Raziskave so opravljali predvsem v laboratorijih Alana R. Fershta, ki je na podlagi barnaze razvil metodo za karakterizacijo prehodov proteinov, ki je znana kot [[phi value analysis]].&lt;br /&gt;

Barnaza deluje kot katalizator na hidrolizo, ki poteka na diribonukleotidnih GpN mestih. Cepitev poteka v dveh korakih in sicer, [[kislinsko-bazni mehanizem]], med prvo [[transesterifikacijo]] se oblikuje ciklični posrednik, ki je kasneje hidroliziran, zato da lahko izloči pritjeno RNA. Dva najpomembnejša ostanka vključena v katalizo sta Glu73 in His102, oba sta prav tako nujna pri encimski aktivnosti. Glu 73 je splošna baza, medtem ko je His102 splošna kislina.
