Najpomembnejši kofaktorji flavoproteinov so torej derivati riboflavinov, ki so sestavljeni iz izoaloksazinskega obroča, na katerega je vezana veriga, ki je derivat sladkornega alkohola ribitola. Obroč je zelo pomembna strukturna komponenta, ki je zaslužna za svetlobno absorpcijo pri UV-žarkih in za rumen izgled flavina in flavoproteinov. Flavin je namreč dobil ime ravno po svoji rumeni barvi, ker se ta v latinščini imenuje &amp;#039;&amp;#039;flavus&amp;#039;&amp;#039;.&lt;br /&gt;

Prisotni so pri mnogih bioloških procesih: pri fotosintezi, popravljanju DNA, apoptozi oz. celičnemu samomoru, metabolizmu in tudi pri bioluminiscenci-oddajanju svetlobe pri živih bitjih, ki nastane pri pretvorbi kemične energije v svetlobno.&lt;br /&gt;
Organizmi dobijo energijo za ta proces pri porabljanju energijsko bogatih molekul. Primer: bakterije porabljajo flavoproteine.&lt;br /&gt;

V znanstveni literaturi so bili prvič omenjeni leta 1879, ko so pri raziskovanju sestave kravjega mleka izolirali svetlo-rumen pigment, imenovan flavin. Kasneje so ta pigment odkrili tudi v vitamin B kompleksu. Dandanes pa je razvitih že mnogo tehnik rekombinantne DNA, ki se ukvarjajo s kloniranjem in karakteriziranjem genov, ki kodirajo flavoproteine, saj je interes po raziskovanju strukture, funkcije in mehanizma le teh zelo velik.
