Heksokinaza je encim, ki fosforilizira šest-ogljikov sladkor, heksozo, nastane hekso fosfat. V večini tkiv in organizmov, je glukoza najpomembnejši substrat heksokinaze, in glukoza-6-fosfat najpomembnejši produkt. &lt;br /&gt;
&lt;br /&gt;
Glukoza je edinstvena v tem, da se lahko uporablja kot vir energije, ki ga vse celice uporabljajo v navzočnosti ali ob odsotnosti molekularnega kisika (O2). Glukoza presnovo preko presnovne poti znana kot glikoliza, je pomembno povezano z mitohondrijskimi oksidativnimi presnovami v prisotnosti kisika, ampak lahko tudi glikoliza ustvarja ATP v odsotnosti. Prvi korak v tem zaporedju reakcij je fosforilacija glukoze, heksokinaza za pripravo na poznejše razdelitve, da se zagotovi energijo.&lt;br /&gt;

Fosforilacija heksoze kot glukoza pogosto omejuje, da bi število znotrajceličnih presnovnih procesov, kot so glikoliza ali sintezo glikogena. Fosforilacija naredi hexose da se ne more premakniti ali odpeljati iz celice.  &lt;br /&gt;

Obstajajo štiri za sesalce pomembne heksokinaza izoencimov, ki se razlikujejo po lokacijah in obmolekulski kinetiki glede na različne podlage in pogoje, ter fiziološke funkcije. Ti so imenovani heksokinaze I, II, III in IV ali heksokinaze  A, B, C in D. &lt;br /&gt;
&lt;br /&gt;
Heksokinaze I, II in III se lahko fizično pridružijo na zunanjo površino zunanje membrane mitohondrija z posebnimi vezavami na Porin ali napetostno odvisnih kanalov anion. To združenje podeljuje heksokinazi neposreden dostop do ATP, ki ga mitohondriji, ki je eden od dveh substratov heksokinaze, proizvaja. Mitohondrijska heksokinaza je zelo povišana v hitro rastočih malignih tumorskih celicicah, z nivoji do 200-krat višje od normalnih tkiv. Mitohondrijskih vezanih heksokinaz je bilo dokazano, da je gonilna sila za izjemno visoko glicolitični tečaji, ki potekajo aerobno v tumorskih celicah (tako imenovane Warburg opisanega učinka).&lt;br /&gt;
