Dva razreda PTK je prisotnih v celicah: transmembranski receptor PTK in nonreceptor PTK.  Fosforilacija proteinov pri kinazah so pomemben mehanizem v pretvorniških signalih za ureditev aktivnost encimov. Približno 2000 kinaz je znanih ,obstaja več kot 100 3D struktur tirozin kinaze (primer je kristalna struktura domene tirozin kinaze v človeškem inzulin receptorju) in več kot 90 protein tirozin kinaz  je bilo odkritih v človeškem genomu. Te igrajo ključno vlogo v različnih celičnih dejavnostih, vključno z rastjo, diferenciacijo, migracijo in metabolizmom in so aktivne pri razvoju organizma in odraslih homeostazah. PTK tudi igra primarno vlogo pri nastopu ali napredovanju bolezenskih stanj, kot so diabetična  retinopatija, ateroskleroza in rak.  &lt;br /&gt;

Encime, ki opravljajo to spremembo so proteini tirozin kinaze, ki katalizirajo prenos fosfata iz ATP v tirozinski ostanek na beljakovini substrata. ki običajno vsebujejo serin, treonin ali ostanke tirozina. &lt;br /&gt;
&lt;br /&gt;
== RECEPTOR ==&lt;br /&gt;
Trenutno je 58 receptorskih tirozin-kinaz (RTK) znanih, združenih v 20 poddružin. RTK so sestavljene iz zunajcelične domene, ki lahko veže poseben ligand, transmembranske domene in znotrajcelično katalitsko domeno, ki s svojimi dejanji zavezuje in fosforilizira izbrane substrate. Vezava liganda na zunajcelični regiji povzroča vrsto strukturnih preureditev v RTK, ki vodijo do njegove encimsko aktivacije. Še posebej pretok nekaterih delov domene kinaze omogoča prost dostop do adenozin trifosfata in substrata na aktivno mesto. To sproži zaporedje dogodkov, s fosforilacijo znotrajceličnih beljakovin, ki v končni fazi prenašajo ekstracelularne signale do jedra, ki povzroča spremembe in genska izražanja. Veliko RTKs je vključenih v onkogenezo, bodisi z gensko mutacijo, ali prenos kromosomov ali preprosto preko izražanja.&lt;br /&gt;
