Vse tri izomere so  vezane na tropomiozinske fibrile, ki uravnavajo krčenje. Troponin C  se nahaja v srčni in prečno progasti mišični celici ter v skeletni celici. Spada pod kategorijo regulatornih proteinov, ki v času mirovanja, takrat ko je koncentracija kalcijevih ionov 10-7   M, preprečuje vezavo miozina in aktina in zato je mišična celica relaksirana. Z dotokom Ca2+  v celico skozi kalcijeve kanale se koncentracija iona dvigne za tri velikostne razrede. Takrat se Ca2+ ioni vežejo na troponin C  in tako povzročijo indirektno kontrakcijo srčnih in skeletnih mišičnih celic. Troponin C je naravni supresor kontrakcije. Kompleks med troponinom C in Ca2+ pa nima več inhibitornega učinka, kar pomeni da lahko miozin in aktin interagirata in posledica je kontrakcija mišične celice.&lt;br /&gt;
