Encim nukleaza S1 je endonukleaza, ki katalizira cepitev fosfodiestrskih vezi med nukleinskimi kislinami. Pogosto se uporablja v laboratoriju pri molekularni biologiji. Njena molska masa je ~31 kDa. Ob določanju aminokslinskega zaporedja nukleaze S1 je bilo potrjeno, da je monomer, torej iz ene polipeptidne verige. Ta je dolga 267 aminokislinskih ostankov z N-glikoziliranim Asn na mestih 92 in 228.
Funkcije nukleaze S1 [1]
- Selektivno cepi enoverižno DNA in RNA, s petkrat večjo aktivnostjo na DNA
- Odceplja enoverižne konce dvoverižni DNA (dsDNA) in tako tvori tope konce (to se rabi npr. pri sintezi cDNA iz mRNA
- Mapiranje transkriptov (identificira 5' in 3' konce gena
- Linearizira dodatno zvite plazmide
- Specifično cepi antikodone tRNA
- Redko cepi tudi dsDNA (dvojna), če so prisotne zareze
Optimalni pH njenega delovanja je med 4.0 – 4.6, aktivira pa se ob prisotnosti Zn2+ ionov [2] (nukleazo S1 izolirano iz Neurospore crassa aktivirajo tudi Ca2+ ioni).
Inhibitorji, ki zavirajo njeno aktivnost so: kovinski kelatorji – EDTA in citrati ter visoke koncentracije NaDS (SDS). Inhibira pa se lahko tudi v nizkih koncentracijah spojin, ki vsebujejo fosfate. Encim se deaktivira pri 10 minutnem segrevanju na 70°C, v prisotnosti EDTA (ta nase veže proste kovinske ione).
Nukleazo S1 večinoma pridobivajo iz plesni Aspergillus Oryzae.
